That will never happen. S-class is an icon, and I guess it will remain sales & image leader in the segment forever. Just like eg. the 3er in it's segment.
The crown of being most comfortable & cozy yet still dynamic luxury sedan? That's S-class domain as well.

But in the "modern & trendy alternative to S-class" department F01 easily beats W221. As the E65 did with W220. But that was not the case with former 7er models (E38, E32) which were only a sportiest & more driver oriented alternative to S-class but not being more trendy & modern.
What E65 lacked was classy appearence - and that has changed already a bit with the FL, but with the F01 can be described as classy again - without compromising the avant-garde sporty dynamic image. Something original E65 failed - since it was avant-garde & innovative + controversial only. Not dynamic enogh, not sporty enough, not classy enough. With F01 these issues were successfully addressed. F01 is way better alternative to S-class any 7er ever was - incl the E65.
But saying that S-class dominance in this segment won't be lost. But be sure F01 will be the most successful 7er up to date.
